摘要

【研究目的】为筛选对苯酚耐受力较高的降解菌,获得降解苯酚的生物材料。【方法】本研究采用苯酚降解菌富集、分离技术从朝鲜蓟分离筛选到一株植物内生细菌菌株CXJ-1。经形态观察、生理生化鉴定及16S rDNA序列分析确定其种属,并采用4-氨基安替比林分光光度法,测定CXJ-1菌株对不同浓度苯酚的降解率。【结果】结果发现,CXJ-1与土生拉乌尔菌 (Raoultella terrigena)(AB680714.1)16S rDNA保守性片段有100%的同源性,可以将该菌株鉴定为土生拉乌尔菌 (R. terrigena)。该菌株对苯酚的最高耐受浓度达2500 mg/L,当苯酚浓度为1500 mg/L时,菌株CXJ-1在48 h内能完全降解。【结论】菌株CXJ-1对由于农药的大量使用而导致苯酚残留的土壤及污水的处理具有较好的应用前景。

Abstract

[Objective] In order to screen out a degrading bacteria that had high tolerance for phenol and degradation of phenol biological materials.[Method] The study separated out a plant endophytic bacteria strains CXJ-1 from Artichoke by adopting Phenol degradation bacteria enrichment and separation technologies.And then determined the species of CXJ-1 through morphology observation, physiological and biochemical identification and 16S rDNA sequence analysis ,measured degradation rate of CXJ -1 strains for different concentrations of phenol by using 4-amino anti than stand light spectrophotometry.[Result] The results of the study found that homologous rate of 16 s rDNA conservative fragment between CXJ-1 strains and Raoultella terrigena was 100%,so The strain could be identified as R. terrigena.Largest concentrations of phenol that The bacteria could toleranced was 2500 mg/L,CXJ-1 Could be completely biodegraded in 48 hours When the concentration of phenol reached 1500 mg/L.[Conclusion] CXJ-1 Had a good application prospect for treatment of Phenol remaining soil and sewage which resulted from extensive use of pesticides.
