金针菇杂交菌株漆酶酶学性质的研究

摘要

为研究金针菇漆酶的酶学性质,试验选取金针菇杂交菌株L7号作为试验材料,对其进行非变性聚丙烯酰胺凝胶电泳(Native-PAGE),发现其含有3种漆酶同工酶,同时采用分光光度法,以ABTS[(2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)]为底物测定金针菇杂交菌株漆酶活性的最佳反应条件。结果表明:适宜测定波长为410 nm、体系pH 4.5、反应温度45℃。

Abstract

Abstract: L7 as experimental material is hybrid strains of Flammulina velutipes. The native—PAGE analysis showed that it possessed three kinds of laccase isoenzymes. By using spectrophotometry and choosing ABTS[2,2’—Azino-bis(3一ethy lbenzothia— z0line一6—sulfonicacid)],the activity of laccase can be determined, the results show that: the appropriate determination of a wavelength 410 nm,system pH4.5, reaction temperature 45℃.
