中秋酥脆枣组培技术研究

摘要

为了建立中秋酥脆枣的快繁体系,本研究以中秋酥脆枣为对象,从外植体消毒时间长短、种类选择、不定芽诱导、不定芽增殖和不定根诱导等方面进行研究,结论是水培枝枣头为最佳外植体,用升汞处理5min后,接种于MS(改良)+6-BA2.5mg/L+NAA 0.15mg/L能获得较高的诱导率, MS(改良)+6-BA1.5mg/L+NAA0.5mg/L是理想的增殖培养基; 1/2MS(改良)+NAA0.15mg/L为理想的生根培养基,成功再生出了中秋酥脆枣。

Abstract

In order to establish the propagation system of‘Zhongqiusucui zao’, the sterilizing time of explants, kind choosing of explants, induction and proliferation of adventitious buds and roots were studied. The result showed that the bud explants from water-cultured branches was the best explant. This kind of explant could be sterilized by 0.1% HgCl2 for 5 min; MS (ameliorated) medium with 2.5 mg/L 6-BA and 0.15 mg/L NAA was the optimum starting medium. The induction rate of sprout was 94%. MS (ameliorated) medium with 1.5 mg/L 6-BA and 0.5 mg/L NAA was the optimum proliferation medium. The sprout number per explant amounted to 6.5. The optimum rooting medium was 1/2 MS (ameliorated) with 0.15 mg/L NAA. Sprout had an average root number of 3.6.
