人工生产僵蚕菌种质量检验

Quality Inspection of Bacteria for Bombyx Batryticatus by Artificial Feeding

摘要:

本研究旨在保障人工养殖僵蚕成功,建立人工生产僵蚕菌种(球孢白僵菌孢子粉)的检验方法和质量评价指标。以不同来源僵蚕菌种为试材,用血球计数板对萌发后的孢子计数,用平板菌落计数法测定杂菌和球孢白僵菌落数。液固双相发酵制备的僵蚕菌种,萌发活孢数和平板活孢数均>1010孢子/g,真菌杂菌数<105 CFU/g,细菌杂菌数小于<104 CFU/g;自然僵蚕菌种萌发活孢数和平板活孢数均<1010孢子/g,真菌杂菌数>105 CFU/g,细菌杂菌数>108 CFU/g。建立的僵蚕菌种检验方法可行,液固双相发酵法制备的僵蚕菌种各项指标均优于自然僵蚕菌种,可在人工僵蚕基地推广应用。

Abstract:

The aims are to ensure the success of cultured bombyx batryticatus by artificial feeding, establish the method and quality assessment index of bacteria for bombyx batryticatus. Bacteria of bombyx batryticatus from different sources were tested. The blood counting chamber was used for counting sprouted spore and flat colony counting method was applied to determine the number of mixed fungi and Beauveria bassiana. Both the number of sprouted active spore and active spore in plate of bombyx batryticatus bacteria made by liquid-solid dual-phase fermentation were greater than 1010 spore/g, while the number of mixed fungi was less than 105 CFU/g, and the number of mixed bacteria was less than 104 CFU/g. Meanwhile, both the number of sprouted active spore and active spore in plate of bombyx batryticatus bacteria produced by nature were less than 1010 spore/g, while the number of mixed fungi was greater than 105 CFU/g. and the number of mixed bacteria was greater than 108 CFU/g. The test method for bombyx batryticatus bacteria is feasible. The index of bombyx batryticatus bacteria made by liquid-solid dual-phase fermentation is better than that by nature, and could be popularized in the culture base of bombyx batryticatus by artificial feeding.
