香子含笑叶挥发油GC-MS分析及抗菌性研究

GC-MS Analysis and Antimicrobial Study of Volatile Oil from Michelia hedyosperma Leaves

摘要: 为研究海南木兰科植物香子含笑的成分及抗菌性,进而为开发植物源防腐剂提供理论依据,该文以生长于海南霸王领区域的香子含笑新鲜叶片为原料,经水蒸气蒸馏法提取其挥发性物质。分离纯化后,采用GC-MS技术进行化学成分分析,共鉴定出34种化合物,占总含量的88.01 %。其中大多为倍半萜类,此外还包括部分单萜类及酸酯类萜的含氧衍生物。鉴定出含量最多的成分为石竹烯(11.442 %)、去氢白菖烯(11.319 %)、3-蒈烯(9.412 %)、香木兰烯(8.981 %)、氧化石竹烯(7.721 %)。通过滤纸片扩散法验证香子含笑叶提取物对大肠杆菌、金黄色葡萄球菌、沙门氏菌生长有不同程度的抑制效果,其MIC分别为0.313、0.625、0.625 g/ml。该研究结果是对国家林木种质资源平台中关于香子含笑成分、功能的补充,也为以该植物叶为原料开发医药、化工、食品等领域新产品提供了理论依据。

Abstract: To study the composition and antibacterial activity of Magnoliaceae plants, volatile oil was extracted by the steam distillation method from fresh leaves of Michelia hedyosperma grown in the Bawangling area of Hainan. After separation and purification, the chemical composition was analyzed by GC-MS and a total of 34 compounds were identified, accounting for 88.01% of the total composition. In addition to oxygen derivatives of monoterpenes and terpenes, most compounds belonged to sesquiterpenes. The components with higher abundance were Caryophyllene (11.442% ), Spathulenol (11.319% ), Carene (9.412% ), Aromadendrene (8.981%), Caryophyllene oxide (7.721%). In this study, the inhibitory effect of extracts from the leaves of M. hedyosperma on the growth of Escherichia coli, Staphylococcus aureus and Salmonella was studied using a filter paper diffusion method. The MIC for the above bacteria was 0.313, 0.625, and 0.625 g/mL, respectively. M. hedyosperma is a unique and endangered plant resource in Hainan. Under the background of building Hainan into an ecologically livable international tourism island, this study provides a valuable basis for conservation breeding and proliferation of M. hedyosperma. In addition, this study adds important information in the National Forest Germplasm Resource Platform regarding the composition and functions of the plant, and provides references for the development of new products in the fields of medicines, chemicals and foods by using the leaves of M. hedyosperma.
