厚朴苗愈伤组织启动培养研究

Study on the callus initiation culture of Magnolia officinlis shoots

摘要: 厚朴资源用途广泛,利用开发多,市场上供不应求。天然厚朴资源稀缺,需营造厚朴人工林,而厚朴种产量少,种子育苗成为瓶颈。本试验研究厚朴苗愈伤组织启动培养,为组织培养快速繁殖体系提供参考。通过完全随机化法试验设计,运用方差分析、LSD多重比较分析,探讨厚朴苗愈伤组织启动培养的最佳培养基类型、最适激素及培养消毒条件。试验表明:B5作基本培养基的培养效果好,若添加较低浓度NAA会促进启动,较高浓度则有利于形成愈伤组织,若添加6-BA 4.0 mg/L有利于培养;对B5添加不同浓度6-BA、2,4-D,愈伤组织分化率最高的组合为:B5 + 6-BA 3.0 mg/L + 2,4-D 3.0 mg/L,芽体分化率最高的组合为:B5 + 6-BA 4.5 mg/L + 2,4-D 2.0 mg/L;对B5添加不同浓度6-BA、NAA,愈伤组织分化率最高的组合为:B5 + 6-BA 4.5 mg/L + NAA 1.0 mg/L,芽体分化率最高的组合为:B5 + 6-BA 5.0 mg/L + NAA 1.0 mg/L。考虑各激素交互作用,综合认为B5 +5.0 mg/L + NAA 1.0 mg/L对启动培养效果最好。

Abstract: The properties of Magnolia officinlis resources are attractive and in high utilization. It is in great demand at the market. Magnolia officinlis in natural are scarce resources, to force the people to create Magnolia officinlis plantation, but the low-yield of the Magnolia officinlis seed will be a bottleneck to seedling. The callus initiation culture of Magnolia officinlis shoots in the test will provide a reference for rapid propagation system of tissue culture. According to analysis and LSD multiple comparison analysis, the best medium, optimal hormone and culture sterilization conditions to start culturing Magnolia officinlis seedlings callus were studied by completely randomized experimental design. It indicated: the basic medium B5 was better, the lower NAA concentration could promote to initiation culture, and the higher was benefical to form callus; when added different (6-BA) and (2,4-D) into medium B5, the differentiation rate of callus was highest in the medium (B5 + 6-BA 3.0 mg/L + 2,4-D 3.0 mg/L), the buds the medium (B5 + 6-BA 4.5 mg/L + 2,4-D 2.0 mg/L); when added 6-BA and NAA into medium B5, the medium (B5 + 6-BA 4.5 mg/L + NAA 1.0 mg/L) was best to differentiate callus, the medium (B5 + 6-BA 5.0 mg/L + NAA 1.0 mg/L) the buds. Considering hormone interaction, the mediume (B5 +5.0 mg/L + NAA 1.0 mg/L) was the best to start culturing Magnolia officinlis seedlings callus. The study will provide a reference for rapid propagation system of Magnolia officinlis seedlings tissue culture.
