Genetic Diversity Analysis of Cymbidium goeringii 's Germplasm Resources Based on SRAP Markers

摘要: 利用SRAP分子标记对42份春兰品种及1份多花兰、1份春剑及3份杂交兰进行遗传多样性分析。筛选出的15对引物组合共扩增出185个位点,其中多态性位点184个,平均每对引物组合产生12.27个多态性位点。引物组合Me8-Em16的Nei’s基因多样性数H(0.3993)、shannon信息指数I值(0.5794)和有效等位基因数Ne(1.7280)都是最高值。47份材料的遗传相似系数变化范围为0.63~0.80,UPGMA 聚类分析表明,在遗传相似系数为0.662处,将47份材料划分成五大类群.本研究较好地揭示了47份材料亲缘关系的远近,为春兰各品种间的分类和杂交亲本的选择提供重要理论依据。

Abstract: SRAP was used to analyse the genetic diversity analysis of 42 Cymbidium goeringii cultivars and 1 specie of Cymbidium floribundum、1 specie of Cymbidum.longibracteatum and 3 species of Hybrid cymbidium. 15 polymorphic primer combinations were screened and a total of 185 DNA loci were amplified,184 of which were polymorphic. The average number of polymorphic DNA loci amplified by each primer was 12.27. Primer combination Me8-Em16’s Nei’s genetic diversity index(H,0.3993) ,Shannon’s information index( I, 0.5794) and Effective number of alleles(Ne, 1.7280) were the highest. The genetic similarity among 47 germplasm ranged from 0.63~0.80. The UPGMA clustering showed that all the tested cultivars and species were classified into five cluster groups with the similarity coeficient of 0.662.The study were well revealed the 47 materials genetic relatives..The findings of this study would provide an important theoretical basis for Cymbidium goeringii classification and parental selection in cross breeding.
