Development of D Ggenome-specific Marker in Oat

Abstract: The research aims to improve the efficiency of molecular breeding and develop D- Genome-specific markers in oat. Firstly, random amplified polymorphic DNA (RAPD) analysis was performed on 9 oat varieties with 300 random 10-based primers. A repetitive sequence of 1287 bp, named OPAB011287 was identified in the D genome of oat. BLAST search revealed that the sequence of OPAB011287 was highly homologous to a part of a retrotransposon. Based on the sequence of OPAB011287, a pair of sequence- characterized amplified region primers (SCAR) was designed which was more convenient and stable. And the results showed that SCAR marker was specific to target oat with D genome, which provided a powerful tool for oat marker assisted breeding and the better service for the breeding of oat.
