    Research on the Rural Human Resources Development Based on the Platform of Imbark Park of Taiwan Farmers

    Development of imbark park of Taiwan farmers provides rural human resources development with a new platform. Imbark park of Taiwan farmers in Fujian seizes opportunities, makes use of advantages, follows the main line of the transformation of scientific and technological achievements, has become the new position of accelerating agriculture extension, new platform of stimulating peasant employment, new way for rural women to achieve self-development, new way to raise farmer organizational level, and new source of developing rural economy. At the same time, continuous improvement of imbark park will be faced with constraints from aspects of policies, systems, markets and so on, which needs government to increase supports, to strengthen training work propaganda, to advance the combined training mode, and to develop new industries to solve problems encountered.

    Hardening Mechanism and the Improvement on the Urban Landscape Greenbelt Soil

    In order to improve the quality of urban landscape greenbelt soil, to keep the urban greenbelt sustainable and healthy, the important characters of urban greenbelts in the nine main districts of Chongqing, such as the bulk density, anthropic-materials,organic matter, available N, P, K and biological quality, were investigated. The results showed that there were five most significant factors resulting in the hardening of the urban soil. The first was the artificial compaction. The total average value of soil bulk density was (1.47±0.05) g/cm3, the highest one was 1.72 g/cm3. The second was a lot of anthropic-materials. The average number was (40.2±4.43)%. The third was low organic content. The average value was (27.2±1.1) g/kg, available N and K content were low, available N content was (39.4±1.1) mg/kg, while available K was (87.3±1.9) mg/kg. The fourth was the unreasonable plant disposition. The last factor was unbalance in the population of soil microbes. The average number of bacteria, fungi and actinomycete respectively were (29.71±7.8)×105 cfu/g, (9.98±1.55)×103 cfu/g and (7.6±1.64)×105 cfu/g. Based on the principles of landscape ecology and symbiosis science, the integrative techniques were put forward for improving hardened and impervious urban landscape soil. The integrative improved techniques will benefit to the sustainable development of the urban greenbelts.

    Agricultural Supply-side Structural Reform: The Connotation, Perspectives and Main Task

    The connotation of agricultural supply-side structural reform is to promote structural adjustment through reform, and to solve the supply problem in structural adjustment way. Its goal is to make quantity, structure and quality of the agricultural supply more adaptable to the natural ecological requirements and more in line with the consumers’demand, with a rational and effective supply of agricultural products. To push forward agricultural supply-side structural reform, the bottom line of grain self-sufficiency and absolute grain ration security must be hold, the transformation of agricultural development mode must be accelerated, the quality, efficiency and competitiveness of agriculture production must be improved, and the market-oriented conception based on all food, whole industry chain and green ecology must be established. Its main task is to reduce inventory, to lower the cost, to bolster areas of weakness, to optimize capacity of foods, to strengthen ecological protection, and to promote the reform. At the same time, the agricultural and rural policy system should be improved, such as financial subsidies, price support, trade protection, finance and insurance services, rural resources and assets management and so on.
    Study on the Functional Activity of Pineapple Polysaccharide in vitro

    Pineapple polysaccharide was extracted from pineapple peel to discuss the antioxidant function of polysaccharide pineapple in vitro. Hot water extraction was used to extract polysaccharide pineapple. The ability of pineapple polysaccharide to scavenge free radicals was studied by using hydroxyl radicals system, superoxide anion system and polysaccharide reductive ability system. Results showed that, using salicylic acid colorimetry to determine the ability of pineapple polysaccharide to scavenge hydroxyl radicals, when the concentration was 12 mg/mL, the scavenge rate reached 90%. While using pyrogallol autoxidation method to determine the ability of pineapple polysaccharide to scavenge the superoxide anion, when the concentration was 6 mg/mL, the clearance of super oxide anion reaches 80%. The reductive ability of pineapple polysaccharide was enhanced with its concentration was increased, and there was a good concentration-response relationship between the concentration and reductive ability.

    The Integrated Technology and Benefit Analysis of Establishing High-yielding Rape in Yunnan

    Based on the definite rape varieties in the project area, according to the higherseedrate, the high cost of field management and low utilization of fertilizer and pesticide of the traditional rape in the production. After many years of accumulated experience, Agricultural Technique Extension Station, Long Yang District has a set of synthesized technology integration of high-efficiency production which includes minimum tillage and no-tillage cultivation,the ditching with machine, suitable sowing time, a one-off thinning and ascertaining quantities seedling, simplifying efficient fertilizer treatments, by unifying management to breeding strong seedlings, machinery prevent aphids ,and so on, and screened two early maturity, high quality, high yield and with stronger drought rape varieties-cloud oil hybrid 2, cloud attar 9, which were extended in the rape production areas and similar climate surrounding earlier maturity rape areas in Yunnan, and have lifted expanding of rape planting area. The project established the high-yielding sample rape area of 680 hectares, the average yield was 3478.5kg/hm2, which increased by 18.6% with compared non-demonstration zone, the average yield increased 545.1kg/hm2, the new total output was 370,500kg, the value was 185.33 million yuan,and reduced expenses of 825yuan/ hm2, rape area of 680 hectares may reduce 56.1 million yuan , by the widespread radiation and driving, the average yield increasing of rapeseed was 150-225kg/hm2, which brought very significant economic benefits and social benefits.

    Application of Tea Polyphenols

    Tea polyphenol is one of the characteristic ingredients in tea, which is about 20%-30% of the total quantity of dry matter. Because of tea polyphenol contains multiple active hydroxyl, which makes it has multiple health care functions such as: antioxidation, anti-aging, sterilize, prevent old cardio-cerebral diseases, which had become the hot spot in medicine, food, the development of animal husbandry etc, and had developed hot tea polyphenols tablet, capsule, weight loss products, beauty products, etc. The antioxidant, sterilizing and anti-aging of activity tea polyphenol so strong that could act as antioxidants in cereals, oils and foodstuffs, aquatic products, meat products, the convenient food had good application aspects. And it also could rise to fight bacterium diminish inflammation, reduce weight, step-down, such as effectiveness could in medicine, hairdressing, etc widely applied. At the same time, it also could use in chemical industry, animal husbandry, etc.

    Breeding of Transgenic Hybrid Cotton Variety‘Yumian 50’and Cultivation Technologies

    The middle-early-maturing new cotton variety‘Yumian 50’was bred by the Institute of Industrial Crops, Henan Academy of Agricultural Sciences. It was examined and approved by Henan Evaluation Committee of Crop Variety in 2016 (Evaluation code: Yushenmian 2016002). In order to study the variety characteristics of‘Yumian 50’and the key points of its cultivation technology, and accelerate its application in cotton production, the breeding process of‘Yumian 50’was described in this paper. The test data of 26 times for three years of‘Yumian 50’participating in Henan regional tests in 2012-2014 were concluded. The results showed that‘Yumian 50’had outstanding performance in Henan regional tests, it was a new cotton variety with high-yield, good quality and disease-pest resistant characteristics.
    Death causes of Chinese pine (Pimus tabulaeformis) at the Tiananmen Square in spring 2010 and technical measures

    In order to explore some causes why pine needles turned yellow and many Chinese pine trees gradually died at Tiananmen Square in Beijing, China in spring 2010, a field investigation was conducted on the external morphology and the root characteristics of Chinese pine, and meanwhile soil physical and chemical properties and site conditions were also investigated and analyzed. It is found that for most of Chinese pines trees, the pine needles of their whole plants turned yellow, their fine roots withered and shriveled, and mycorhiza were rotten and died. The high salt content was found in surface soils, and the roots absorbed the large amounts of salts and lost the function of absorbing water and nutrients, which led to the emergence of yellow pine needles and the death of the whole plant. Some factors (such as small growth space, high soil density, poor soil ventilation and heat radiation) were not in favour of normal growth of Chinese pine. Moreover, a great amount of snow-melting agent was applied around Chinese pine at the Tiananmen Square after snow in winter 2009, which was a direct cause leading to the death of Chinese pine. In order to protect the achievement in afforestation and visual effects, the corresponding technical measures and suggestions were presented to prevent the hazards of snow-melting agent and to overcome the current status of adverse site conditions.

    Optimization of Western Blot Condition for Detection of Protein Expression in Rabbit Artery

    Western blot is the most important method on studying and analyzing protein. Because it contains many steps, high background, unspecific bands, streaking and so on are the common problems in Western blot, which will influence the subsequent detection and quantitation of protein. To establish the best experiment condition for detection of protein expression in rabbit artery, VCAM-1(vascular cell adhension molecule-1) was detected. A rapid, specific and sensitive procedure has been successfully established, through optimizing several conditions which include SDS-PAGE, antibody dilution, temperature and time for antibody incubation, detection methods and so on. Also, three kinds of detection methods were used which contain DAB, enhanced chemilluminescence (ECL) with exposure captured on X-ray film or with cooled charged-coupled device (CCD) cameras. The sensitivity of three methods was compared. The results indicated that ECL has sensitivity 2-10 times that of DAB, and CCD cameras have 4 times sensitivity of X-ray and a more precise procedure. So, ECL-CCD cameras is a promising detection method of Western blot.

    Effect of Wilt and Additives on Red Clover Silage

    The purpose of this paper was to investigate the effects of different additives on fresh or wilted red clover silage. The red clover during full flowering stage was moved and ensilaged in bags. Three additive treatments were designed. They were control without additive, formic acid (6 mL/kg silage) and sucrose (2%). The silage was sampled and analyzed after 360 days of storage. The fermentation quality of silage was improved in the formic acid or the sucrose treatment. The pH of red clover silage was extremely significantly improved under wilt conditions (P<0.01), but lactic acid was extremely significantly decreased (P<0.01). The pH and the contents of acetic acid and propionic acid in the red clover silage were extremely significantly decreased in the formic acid or the sucrose treatment (P<0.01), but the lactic acid was extremely significantly increased (P<0.01). The dry matter recovery of silage was significantly increased in the formic acid treatment (P<0.05). The nitrate content of red clover was reduced after being ensilaged.

    Effect on Yield of Super Rice‘Liangyou 616’: Seedling Age, Transplanting Density and Nitrogen Application Rate

    To reveal the integrative effects of seedling age, transplanting density and nitrogen application rate on yield of middle-season rice‘Liangyou 616’, and study its high-yield cultivation technique, an experiment was designed by using the 311-A optimum regression, and a mathematics (regression) model was established between yield and the factors, seedling age, transplanting density and nitrogen application rate in the filed experiment. The results indicated that the effect of the three factors on yield was as following: nitrogen application rate >transplanting density > seedling age. With the simulation and optimization by computer, the results suggested that the optimal combination was seedling age of 30.4d, transplanting density of 204600 clump/hm2,and net N fertilizer of 168.75 kg/hm2, and the imitated highest yield was 9778.1 kg/hm2. The main agronomic measures of‘Liangyou 616’to achieving the yield of more than 8625 kg/hm2 were seedling age of 29-33 d, transplanting density of 172000-227000 clump/hm2 and net N application rate of 160.0-190.8 kg/hm2.
    Effect of Waterlogging Degree on Cotton Seedling Growth and Physiological Change

    In order to clarify the adaptability of cotton after waterlogging stres. cotton morphology, production and physiological changes at the seedling stage under different flooding degree were studied by using a pot experiment. The results showed that after flooding, cotton production, the rate of bolls per plant, boll weight, plant height, number of leaves and dry matter accumulation of per plant reduced, while Lint percentage did not change; with the flooding time increasing, various indicators continued reducing. after flooding 20d, leaf SPAD value declined rapidly, cotton lint yield decreased by more than 80%, even total crop failure. Overall, flooding 15d could be considered the critical time of cotton to adapt waterlogging stress during seedling stage , so cotton can resuming growing through cultural technique measure before the critical time.
    Analysis on the Bioinformatics of Chalcone Synthetase Gene in Petunia hybrida Vilm.

    To study the homology and evolutionary relations among CHS (chalcone synthase) genes, CHS gene in Petunia hybrida, Vilm. was took as explants material, base distribution, amino acids composition, prediction of hydrophobicity or hydrophilicity, and multiple alignments with other 10 species were analyzed by the software and bioinformatics web site. The results showed that the mRNA single sequence contained 1170 bp base, the number of amino acids encoding protein was 389 and its relative molecular mass was 42580.02, Leucine achieved the highest content of 10.80%, hydrophobic maximum value was 2.038, hydrophobic minimum value was -2.208, and no obvious hydrophilic or hydrophobic area. The homology of multiple alignment of the amino acid sequence for CHS genes in 11 species achieved 80.55%. The results showed that the genes were in a stable state, encoded proteins were hydrophilic protein, and the evolutionary process was conservative. The conservative sequence information achieved in these CHS genes laid a foundation for other new CHS gene cloning.

    Planning on the Construction of Chang guangxi Wetland Park in Wuxi

    Besides the wetland reserves, the wetland park is an important component of wetland conservation system, and it is a management approach that is adopted to protect the natural or artificial wetland which is affected by human beings but still has its conservation value. In order to make healthy and orderly progress of wetland parks in China, some ideas are presented and discussed in this paper using the Chang Guangxi Wetland Park in Wuxi, Jiangsu Province, China as the study field. They are function zoning and general layout of the wetland Park, wetland park landscape construction, publicity and education engineering construction, scientific research monitoring, tourism infrastructure construction, health and safety engineering construction, park’s staging construction, construction management, etc.

    The Use of Eugenol as an Anaesthetic for the Australian Longfinned Eel, Anguilla reinhardtii (Steindachner)

    Anaesthesia is important in handling large river eels during procedures such as measuring, tagging and surgery. Therefore, the use of eugenol as an anaesthetic for the Australian longfinned eel, Anguilla reinhardtii (Steindachner) was investigated with a range of concentrations (10-120 mg/L). 20-120 mg/L of eugenol was found to be safe and effective (P <0.05), which could induce anaesthesia in less than 10 min, and recover after anaesthesia also in less than 10 min with a recovery rate of 100%. The difference of induction times and recovery times among 40, 60, 80 and 100 mg/L was not statistically significant, but with and among other groups was significant (P <0.05). Interestingly, average induction times did not always drop as the concentration increased. The induction time decreased from 312.88 s to 252.25 s when the concentration increased from 20 mg/L to 30 mg/L, while increased greatly to 582.00 s when attained 40 mg/L. Between 40-120 mg/L, induction times were inversely correlated with concentrations (r =-0.84, P <0.05), and dropped again in general along with the increase of concentration. Mean recovery time was the shortest for 20 and 30 mg/L, and shorter for 120 mg/L, and showed correlations of a certain degree both with anaesthetic concentration and induction time (r = 0.52, P <0.05). In conclusion, eugenol is a safe and effective anaesthetic for the Australian longfinned eel. 20-30 mg/L of anaesthetic concentration which can induce anaesthesia in 4-5 min in average and recover after anaesthesia in 4-7 min is recommended.
    Biological Characteristics of Begonia fimbristipulata Hance.

    To accurately master the biological characteristics of Begoniaceae Begonia fimbristipulata Hance., and to better protect the unique local plant resources, further research and additional revise have been made on this species' six features, such as generic names, botany forms, environmental growing distribution, growing habits, collecting process and medicinal functions, through years of field investigation, domestication, cultivation of Begonia fimbristipulata Hance. in Dinghushan Mountain. The biological data of Begonia fimbristipulata Hance. has been systematically perfected in order to provide scientific evidence for this species' protection and correct use.

    Research Progress of Heavy Metal Contamination in Soil and Remediation Method

    In this paper, the current situation of heavy metal pollution in soil was discussed in such aspects of major form of heavy metal in soil, assessment index of pollution level, circulation mechanism, mutual effect of ions and remediation methods in order to study the research development of different remediation methods and the future works of heavy metal pollution. The future of research status was prospected in an effort to provide reference for other researchers. Results showed that basic research of heavy metal pollution in soil needed to be further studied, especially the circulation mechanism. Plant and microorganism remediation methods were main parts of heavy metal remediation, together with other remediation methods.

    Analysis on Characters and Yield of Different Maize Varieties

    In order to select the new maize cultivar for Dingxing County, Hebei Province, the comparative analysis on characters and the yield of 7 maize cultivars had been studied under the specific climate and the soil conditions. The results showed that: ‘Jingdan28’ leaf blade had the highest photosynthetic rate and a good comprehensive character. The per unit yield reached 10321.5 kg/hm2. And the yield components of maize were 74025 spikes per hm2, 395 grains per spike, and the 1000 grain weight was 353.0 g. In the 7 tested maize cultivars, the yield of ‘Jingdan28’ and ‘Zhengdan958’ compared with other 5 maize cultivars were extremely different significance. ‘Jingdan28’ and ‘Zhengdan958’ were suitable for Dingxing County and other area with the close light and hot resources for realizing high-yield cultivation and increasing yield, which was of great significance.

    Genetic Diversity Analysis of Wheat Aphids Resistant Wheat Varieties by Simple Sequence Repeat (SSR) Markers

    In order to gain knowledge of genetic diversity of wheat varieties/lines resistance to wheat aphids, on the basis of field insect plot appraisal, 47 wheat varieties/lines with different degrees of resistance to wheat aphids were studied by 18 SSR markers to explore genetic diversity of wheat aphids. 18 SSR markers were screened and a total of 99 allelic variants were detected in 47 wheat varieties ranged from 1 to 10 alleles per marker with an average of 5.21. The mean of alleles of high resistance insect (HR) wheat varieties were 3.83, of resistance insect (R) wheat varieties were 3.88, of sense insect (S) wheat varieties were 4.06 and of high sense insect (HS) wheat varieties were 3.56. The PIC (Polymorphism information contents, PIC) for each polymorphic primer varied from 0 to 0.91 with an average of 0.64. PIC of the HR wheat varieties were an average of 0.60 (0-0.97), of the R wheat varieties were an average of 0.64 (0-0.99), of the S wheat varieties were an average of 0.64 (0.14-0.85) and of the HS wheat varieties were an average of 0.67 (0-0.81). The mean genetic distance (GD) between 47 varieties was 0.65 varied from 0.33 to 0.94. The GD between HR variety was 0.59 varied from 0.39 to 0.77; The GD between R variety was 0.62 varied from 0.39 to 0.83. The GD between S varieties was 0.62, varied from 0.37 to 0.83. The GD between HS variety was 0.64, varied from 0.37 to 0.84. Based on 0.67 of GD, 47 varieties were clustered into seven groups. Resistant insect wheat variety ‘Linyuan 95-5322’ was a groups by oneself. It was explained that there was a further genetic relationship between ‘Linyuan 95-5322’ and other variety, and ‘Linyuan 95-5322’ could be used to resistant insect parent.

    Genomic DNA Extraction of Three Kinds of Fruit Flies and their ISSR-PCR Reaction System Establishment

    The author took three kinds of fruit flies for materials, discussed the extraction of genomic DNA with high quality, studied on the effect of template DNA concentration, primer concentration, the amount of Taq DNA polymerase, dNTP concentration, annealing temperature and time on ISSR-PCR amplified results, established fruit flies’ general and stable ISSR-PCR reaction system. The result showed that the author obtained the fruit flies’ genomic DNA with high quality, established fruit flies’ general and stable ISSR-PCR reaction system that adapt to fruit flies: 2.5 μL 10×PCR Buffer, 50ng template DNA, 0.25 μmol/L primer, 0.5 U Taq DNA polymerase, 200 μmol/L dNTP and add ddH2O to 25 μL, made clear the ISSR-PCR amplification program: pre-denaturalized at 94℃ for 5 min, denaturalized at 94℃ for 30 s, annealed at 52.4℃ for 45 s, extended at 72℃ for 90 s, 36 cycles, at last extended at 72℃ for 7 min, then conserved at 4℃. The establishment of optimized system made up for the shortage of fruit flies’ traditional morphology observation, and laid a foundation for fast and accurate identification, population heterogeneity and analysis of genetic diversity.
