Comparison of Several Methods in Genomic DNA Extraction from Rose rugosa

Abstract: In order to find a convenient, fast and cost-effective genomic DNA extraction method and lay a good foundation for the study of Rose rugosa, its genomic DNA was extracted from young leaves of different varieties of Rose rugosa with CTAB method, SDS method, modified CTAB method and DNA minprep kit method. The quality and quantity of the extracted DNA was tested and compared by means of agarose gel electrophoresis and UV specterphotometry. The results showed that DNA could be extracted with all the four methods, but as an improved method, modified CTAB method could extract DNA of the best quality and the highest concentration under the premise of being economical and practical. However, DNA minprep kit method was a new method of DNA rapid extraction. This method had disadvantages of lower yield and higher cost, but had advantages of simpler operation, shorter time, lower toxicity, higher efficiency and faster. A suitable method could be chosen according to the actual conditions of the labs.
