    The Use of Eugenol as an Anaesthetic for the Australian Longfinned Eel, Anguilla reinhardtii (Steindachner)

    Anaesthesia is important in handling large river eels during procedures such as measuring, tagging and surgery. Therefore, the use of eugenol as an anaesthetic for the Australian longfinned eel, Anguilla reinhardtii (Steindachner) was investigated with a range of concentrations (10-120 mg/L). 20-120 mg/L of eugenol was found to be safe and effective (P <0.05), which could induce anaesthesia in less than 10 min, and recover after anaesthesia also in less than 10 min with a recovery rate of 100%. The difference of induction times and recovery times among 40, 60, 80 and 100 mg/L was not statistically significant, but with and among other groups was significant (P <0.05). Interestingly, average induction times did not always drop as the concentration increased. The induction time decreased from 312.88 s to 252.25 s when the concentration increased from 20 mg/L to 30 mg/L, while increased greatly to 582.00 s when attained 40 mg/L. Between 40-120 mg/L, induction times were inversely correlated with concentrations (r =-0.84, P <0.05), and dropped again in general along with the increase of concentration. Mean recovery time was the shortest for 20 and 30 mg/L, and shorter for 120 mg/L, and showed correlations of a certain degree both with anaesthetic concentration and induction time (r = 0.52, P <0.05). In conclusion, eugenol is a safe and effective anaesthetic for the Australian longfinned eel. 20-30 mg/L of anaesthetic concentration which can induce anaesthesia in 4-5 min in average and recover after anaesthesia in 4-7 min is recommended.
    Immunostimulatory Effects of LPS and β-glucan on Subunit Vaccine of Edwardsiella tarda

    In order to investigate the immunostimulatory effects of LPS and β-glucan, an highly effective vaccine candidate for Edwardsiella tarda known as glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was recombinantly expressed in Escherichia coli, which was injected into Japanese flounder alone or containing LPS and β-glucan. After immunization, the antibody titer, expression of immune-related genes and immunological protection were detected. ELISA analysis of sera showed that the fish immunized with rGAPDH containing LPS and β-glucan had significant higher antibody titers than rGAPDH immunization group at day 14 and 30, which reached a value of 211.76 at day 30 after immunization. Simultaneously, the expression of 6 immune-related genes involved in innate and acquired responses, including IL-1β, IL-6, MHC-IIα, IgM, TCRα and C-type lysozyme were analyzed by quantitative PCR. The transcript levels of all selected genes displayed up-regulation after immunization, and immunization with rGAPDH containing LPS and β-glucan could induce significant higher levels than that induced only by rGAPDH. Moreover, to investigate the protection against infection, challenge was performed at day 30 after immunization. Fish immunized with rGAPDH containing LPS and β-glucan exhibited the highest relative percentage survival (RPS) of 67 % compared to the other groups. These results indicated that LPS and β-glucan employed as immunostimulatory molecules could significantly trigger the innate and acquired immune responses in Japanese flounder.
    Review of High-throughput Sequencing Techniques in Studies of constructedwetland Microbial Diversity

    The paper aims to explain the theory of purifying the water body with wetland from the perspective of microbial diversity. The process and principle of the next- generation sequencing technology—highthroughput sequencing technology was analyzed, the advantages and disadvantages of high- throughput sequencing technology applications and the high-throughput sequencing technology application in wetland microbial diversity were summarized in this paper. Meanwhile the broad application value of high-throughput sequencing technology in the diversity and dynamics of the microbial community of artificial wetland was obtained, the high-throughput sequencing results in huge data processing and other issues were pointed out in this paper. Because the high- throughput sequencing technology has unique advantages, it has a great application prospect in the study of wetland microbial diversity.
    A New Species Sinocyclocheilus longshanensis sp. nov. from Yunnan

    This study elaborates a new species in a karst cave near the foot of Longshan Mountain, Xiaoxinzhai Village, Shupi Township, Qiubei County, Wenshan Zhuang and Miao Autonomous Prefecture, Yunnan Province, China. The new species is a member of Sinocyclocheilus, and named as Sinocyclocheilus longshanensis Li et Wu sp. nov. according to the name of collection region. Morphological observation shows that the new species has 7 branched dorsal-fin rays, trailing edges of the last unbranched dorsal-fin ray is serration; normal eyes; sub-upper mouth; humped back; black-brown round spots distributed on exposed body surface; 59~62 lateral line pores. The new species can be distinguished from Sinocyclocheilus maculayus, which has humped back; black-brown round spots distributed on exposed body surface; 81~88 lateral line pores. The new species can be distinguished from Sinocyclocheilus yangzongensis, which has flat back; golden body color; 71~81 lateral line pores. The new species also can be distinguished from other species of Sinocyclocheilus by having the described combination of morphological characteristics.
    Effects of Fertilizers Application Rates on Growth and Yield of Houttuynia cordata Thunb

    Single factor randomized block design, with the measurement height, branches and fresh weight, was employed to analysis the effects of 5 fertilizers on the growth and yield of Houttuynia cordata Thunb. The result showed that the plant grew slowly at the early stage after transplanted, the application of organic fertilizers was beneficial to the development of plant height and branches at later stage, and manure of duck acted as the best one. It also showed that the application of organic fertilizers increased the yield of Houttuynia cordata Thunb, with a highest yield was detected in application of duck excretion at moderate level, especially for the underground part of the plant, which was higher than the application of chemical and traditional fertilizer. From the result of the test, we suggested that duck excrement was the best one of organic fertilizer for the cultivation of Houttuynia cordata Thunb, and can be mixed with other organic fertilizers, such as manure of human and pig, incooperation with and grow moreover a small amount of quick-acting chemical fertilizer is needed at earlier and later growth stage of the plant. In the application of organic fertilizers, 108000 kg/hm2 of human manure, 54000 kg/hm2of pig manure and 45000 kg/hm2 of duck manure were recommended respectively.
    Investigation of Background Concentration of Formaldehyde in Fresh Fishery Products

    The concentration of formaldehyde in 1013 fishery product samples, which including 314 samples of 12 species of freshwater fishes, 356 samples of 19 species of marine fishes, 147 samples of 9 species of crustaceans, 137 samples of 5 species of shellfish and 59 samples of 4 species of cephalopods, were determined by acetylacetone spectrophotometric method. The results showed that the median of formaldehyde content in fresh fishery products is 0.25 mg/kg, which was far below the average concerntration (6.31 mg/kg), indicating that most samples have low formaldehyde contentration. In the survey of 49 species, bombay duck(Hacpodon neheceus), cod(Cadous macrocephalus, Theragra chalcogramma and Miciomesistius poutassou), squid (Loligo chinensis), squill (Oratosquilla) and swimming crab (Portunus trituberculatus and Portunus pelagicus) had higher levels of formaldehyde in the background. While formaldehyde content in tilapia seemed to be significantly higher than other freshwater fish species (p<0.05). Significant differences on types showed that the formaldehyde conerntration in marine fish samples were great, followed by samples of cephalopods, crustaceans and shellfish, however, samples of freshwater fish was in the lowest formaldehyde content (p<0.05).

    Adding different quantity of rare earth nitrate to the Carassius auratus feed and researching the effect of rare earth on the production performance and nutritional status of Carassius auratus. The result indicates that the effect of adding rare earth nitrate 45mg/kg in the feed was the most obvious, the growth velocity was improved, the feed coefficient was greatly decreased significantly(p﹤0.01), the disease resistance ability was enhanced and the mortality rate was reduced, meanwhile the contents of protein and fat in bodies of fish was obviously improved(p﹤0.05).

    With regard to the biodiversity of living things, FishBase is one of the largest databases in scope and extension .Up to now, FishBase has collected information on 31 000 fish species, covering their biology, ecology, taxonomy, population dynamics, genetics, physiological characteristics and economic values, bearing a high scientific and practical value. China became the ninth consortium member in 2007. The main function and structure of FishBase are summarized and analyzed in this paper. Different retrieval ways are also introduced, with an aim to assist research institutes and related enterprises with retrieval techniques and to provide technological support for the aquaculture industry. It is expected that the database would be better and more effectively used by research institutes and enterprises.
    A preliminary study of rising temperature on immune-related enzyme and MDA content in family of sea urchins(Strongylocentrotus intermedius) with different color tube feet and red, white Strongylocentrotus intermedius

    The experiment studied the change of immune-related enzyme and MDA content in rising temperature. The activities of SOD, CAT, POD, LZM and the content of MDA in family of sea urchins (Strongylocentrotus intermedius) with different color tube feet were analyzed. The result show that:(1)In rising temperature, the change of SOD,CAT,POD,LZM were trend to increased and to decreased in RR, RW, WR, WW family of sea urchins, the activities of SOD were 2.39 ~4.36U/ml, 2.79 ~5.17U/ml, 2.71 ~4.97U/ml,3.66 ~5.69U/ml respectively; the activities of CAT were 63.02 ~112.76U/ml, 62.47 ~169.37U/ml, 66.40 ~140.77U/ml, 72.29 ~149.20U/ml respectively; the activities of POD were 1.65 ~14.59U/ml, 0.89 ~13.88U/ml, 2.08 ~14.68U/ml, 2.49 ~15.62U/ml respectively; the activities of LZM were 9.82 ~109.23U/ml, 12.10 ~110.37U/ml, 7.82 ~112.16U/ml, 11.18 ~107.83U/ml respectively; the activity of MDA first reduced and then increased in RR, RW and WR family, the content of MDA were 0.97 ~2.34nmol/ml, 0.82 ~2.40nmol/ml, 0.96 ~2.23nmol/ml, the activity of MDA in WW family was trend to increased, the content of MDA was 1.55 ~2.60nmol/ml. (2)The change of SOD,CAT,POD,LZM were trend to increased and to decreased in red and white tube feet S. intermedius, the activity of MDA first reduced and then increased in red tube feet sea urchins, and increased in white tube feet sea urchins. (3) In rising temperature, the activity of SOD, CAT in RR family was more sensitive than WW family, the content of MDA in RR family was lower than in WW family; the activity of SOD, POD in red tube feet sea urchins was more sensitive than in white tube feet sea urchins(P<0.05); in 26℃ the activity of POD had significant difference(P<0.05). These result revealed that family of sea urchins presented in the aspect temperature sensitively and adaptability strongly whose foster parents were the red tube feet sea urchins.
    Construction of Plant Expression Vector Containing Genes Encoding The Key Biosynthesis Enzyme of Astaxanthin( bkt) and Its Expression in Zea mays L.

    Astaxanthin with high antioxidant activity is of great application value. Bkt is the key biosynthesis enzyme of astaxanthin which encoded by bkt gene. Bkt gene expression cassette was constructed by replacing GUS gene in vector pBI221 with bkt gene obtained by PCR method, which contained the CaMV 35S promoter and NOS terminator. Plant expression vector pCAMBIA1301-bkt was constructed by inserting bkt gene expression cassette into multiple cloning site in vector pCAMBIA1301.Bkt gene was transferred into maize Qi 319 mediated by Agrobacterium tumefaciens. The results of GUS histochemical assay indicated that bkt gene had been transferred into maize emberyogenic calli.
    Correlation between Meat Yield and Body Measurements in Procambarus clarkii and Cherax quadricarinatus

    This paper provided a measure to estimate the meat yield and to selective breed of Procambarus clarkii and Cherax quadricarinatus. The correlation between meat yield and body measurements in these two kinds of crayfish were studied by statistics analysis. The results showed that, the meat yield value of Cherax quadricarinatus was higher than that of Procambarus clarkii. First abdominal segment width was the main variable to Procambarus clarkii. Body length and carapace length were the main variables to Cherax quadricarinatus, followed by total length. It was concluded that: (1) the correlation to meat yield was more significant than that to body weight; (2) it was possible to predicting meat yield based on ratio of two body measurements or the individual variable in Procambarus clarkii and Cherax quadricarinatus.

    Comparative Analysis of Big City Residents’ Consumption Level and Consumption Characteristics of Aquatic Products in China

    Aqua-products consumption in our country varies greatly in different areas with distinctive natural conditions and unique customs and culture. This paper bases on the questionnaire research in three areas of Beijing, Shanghai and Xi’an. From the perspective of region, it targets on urban residents in different areas to analyze their consumption level, purchasing tendency, consumption variety, main consumption form, dominated trade location and preferences for seawater or fresh water products and captured or farmed ones. Finally, the paper proposes some instructive suggestions to improve the consumption level of aqua-products in different areas.
    Correlation and Path Analysis of Morphometric Attributes of Synodus macrops

    In order to research the correlation of morphometric attributes of Synodus macrops and the effects on the body weight, 150 Synodus macrops were collected and the attributes including body length (X1), body width (X2), body depth (X3), head length (X4) and weight (Y) were measured and analyzed by correlation analysis and path analysis. The results showed that, all correlation coefficients between morphometric attributes and weight achieved very significant difference (P<0.01). The path coefficient of body length (X1), body width (X2), body depth (X3) to the body weight all achieved significant level (P<0.01). These morphometric attributes were the most important determination to body weight. Among them body length had the largest direct effect on body weight, and body depth and body width ranked in order. The head length affected body weight indirectly. The diversification of decision coefficients analysis was consisted with the path analysis. Eliminating the head length which had no significantly direct effect on body weight, the multiple regression equation was constructed: Y=-50.198+0.299X1+0.695X2+1.238X3(R2=0.930).
    Analysis and Evaluation on the Nutritional Components of Acipenser schrenckii

    It is the research on main nutrient components and chemical protein evaluation of Acipenser schrenckii roe that provide reference for deep processing of Acipenser schrenckii roe and scientific basis for exploitation, and comprehensive utilization of Acipenser schrenckii resources. The nutritive components of Acipenser schrenckii roe were determinated by conventional analytical methods. The results showed that the crude protein content of Acipenser schrenckii roe was 26.00%. The 18 amino acids content of Acipenser schrenckii roe was 25.21%. Among these amino acids, the essential amino acids content was 10.16%, and the fraction of essential amino acids was approximately 40.30%. The ratio of essential amino acids to non-essential amino acids was 67.51%. Tryptophane, the first limiting amino acids, scores 92. The unsaturated fatty acids amount of ASO was 62.8%. The EPA and DHA amount of Acipenser schrenckii roe reached to 10.3%. The content of mineral elements, such as Fe and Zn, were high. Its content was 22.07 mg/kg, 18.00 mg/kg, respectively. Acipenser schrenckii roe had the characteristics of high protein, low carbohydrate, and reasonable proportion of amino acid, abundant minerals and abundant unsaturated fatty acid, which had a bright application future.

    A Preliminary Study on Chromosome Preparaation and Karytype of Leptobotia taeniops

    To analysis the chromosome number and karyotype of Leptobotia taeniops. The karyotype of Leptobotia taeniops was studied through the chromosome preparations obtained from head kidney by the method of injecting PHA and colchicines and used air drying method to make chromosome samples. The total chromosome number of the fish was 50, the karyotype formula was 2n=6m+10sm+8st+26t, and NF=66. There was no visible evidence of po1yploidy chromosome and sex chromosome. This study provided Leptobotia taeniops with cytogenetics paramete, and offered a theoretical basis for Cross breeding of Leptobotia taeniops.

    Morphometric Differences Analysis of Pelteobagrus vachelli, Pelteobagrus fulvidraco, Leiovassis crassilabrus and Their Hybrid F1

    In order to provide more discriminant basis for Bagridae fish breeding, through the form and framework data determination of Hybrid groupⅠ(Pelteobagrus vachelli, Pelteobagrus fulvidraco and their positive and negative hybrid F1 generation), and Hybrid groupⅡ(Pelteobagrus vachelli, Pelteobagrus fu lvidraco, Leiovassis crassilabrus, Pelteobagrus fulvidraco♀×Leiovassis crassilabrus♂ F1 generation, and Pelteobagrus vachelli♀×Leiovassis crassilabrus♂ F1 generation), two methods of clustering and discriminant were analyzed, clustering results showed that hybrid F1 generations showed some of the maternal effects in GroupⅠ; in GroupⅡ, Pelteobagrus fulvidraco♀×Leiovassis crassilabrus♂ F1 generation showed more similar to its mother and Pelteobagrus vachelli♀×Leiovassis crassilabrus♂ F1 generation tend to be more familiar to its father. Discriminant anlysise results showed: the GroupⅠpositive and negative hybrid F1 generations showed evenly to their father and mother features, no more apparently showed male or female characteristic; In GroupⅡ, 5 population discriminant accuracy rate was 95.33%, clearly showed the difference between each group morphology. The conclusion: (1) The positive and negative hybrid generations of Pelteobagrus vachelli and Pelteobagrus fulvidraco, Pelteobagrus vachelli♀×Leiovassis crassilabrus♂ F1 generation represent significant maternal effects; well Pelteobagrus vachelli♀×Leiovassis crassilabrus♂ F1 generation represent more paternal effects; (2) Comprehensive use of proportion of trait parameters and calibration framework parameters had more ability than used alone to improve discrimination ability of shape classification. The more detailed parameters, the stronger the discriminant ability.
    Path Analysis for Growth-related Traits of GIFT Strain of Nile Tilapia (Oreochromis niloticus) under Different Stocking Densities

    Four kinds of growth- related traits, which were body length, total length, body height and body width, were selected to study their effects on body weight of GIFT strain of Nile tilapia under different stocking densities. One way ANOVA was applied to analyze the differences of growth- related traits under different stocking densities. Path analysis was employed to study the importance of all variables. After removing variables without significant differences, the results showed that most of the growth-related traits under the three stocking densities were significantly different (P<0.01) except body length and the total length under the densities of 18000 fish/hm2 and 22500 fish/hm2 (P>0.05). Body weight of GIFT strain of Nile tilapia was mostly affected by body length, body width, and total length when the stocking densities were 18000 fish/hm2, 22500 fish/hm2, and 27000 fish/hm2, respectively. So in the regression equations, the variables without significant difference were also removed. They were the trait of total length at the density of 22500 fish /hm2, the trait of body length at the densities of 18000 fish /hm2 and 27000 fish /hm2. To sum up, effects of growthrelated traits on body weight were significantly different under different stocking densities of GIFT strain of Nile tilapia, and should be considered in breeding.
    Bibliometrics Analysis on Hot-topic Fishery Sci-Tech Field Based on Journal Papers

    By analyzing the published papers on JORUNAL OF FISHERIES OF CHINA and JOURNAL OF FISHERY SCIENCES OF CHINA, which are two important academic journals in fishery science in China with high influence, we classified the papers according to different subjects and compared them in terms of paper numbers, and discussed the hot-topic subjects based on the results. The results showed that there were total 1186 papers published on the two journals from 2002 to 2006, among which the papers in the subject of genetic breeding in aquaculture organisms had the highest number, which was 290, making up 24.5% of the total paper numbers. In fact, this subject has been being the hot topic during recent years, and the research papers have been published not only in China but also in some international journals. But for the subject of fishery environment and ecology, the authors had more journals to contribute, so the situation reflected from the two journals may not be general. Anyway, the results of this study can reflect the comparative advantage and disadvantage among the different subjects in fishery science, and the purpose of this study is to provide a reference for the academic journal editors to select their candidate papers and for the researchers to project their researches.
    Effects of Flavomycin on Activities of Digestive Enzymes in Cyprinus Carpio

    An experiment was conducted to investigate the effects of different levels of Flavomycin supplement on growth performance and activities of digestive enzymes in Cyprinus carpio. The results showed: The biology synthesis evaluates in the groups fed diets with Flayomycin were higher and that (119) in the group fed diets with 6mg/kg Flayomycin was the highest. Feed coefficients in the groups fed diets with 4mg/kg and 6mg/kg Flayomycin were significantly higher than that in the control group (P<0.05). The activities of trypsin and intestinal protease were the highest in the group fed diets with 4mg/kg Flayomycin. The activities of trypsin in the group fed diets with 4mg/kg Flayomycin were significantly higher than those in the control group and the group fed diets with 8mg/kg Flayomycin (P<0.05). The activities of intestinal protease in the group fed diets with 4mg/kg and 6mg/kg Flayomycin were significantly higher than those in the control group (P<0.05). The amylase activities in hepatopancreas in the groups fed diets with 2mg/kg, 4mg/kg, 6mg/kg Flayomycin were significantly higher than those in the control group (P<0.05). The amylase activities in intestine in the group fed diets with 4mg/kg Flayomycin were significantly higher than those in the control group (P<0.05). The lipase activities in hepatopancreas and intestine in the group fed diets with 6mg/kg Flayomycin were significantly higher than those in the control group (P<0.05).

    AFLP(Amplified Fragment Length Polymorphism) analysis system for Oncorhyrchus masou masou were established in this study, the genomic DNA were digested by two groups of restriction enzymes, with the relative processes being optimized, including the quantity of the restricting enzymes, the time of digested reaction, quantity of the template for pre-amplification and times of pre-amplification products dilution. It was proved that the results were best with 100ng DNA digested for 3h at 37℃ by 3U EcoRI and then 4h at 65℃ by TaqI and Tru9I respectively, then ligated by ligase. Quite clearly results could be achieved by 3μL ligation products as the template for pre-amplification which products were diluted 50 times for selective amplification. The comparison of the two pairs showed that the group of EcoRI-Tru9I obtained more amplification bands but lower ratio of polymorphic loci than EcoRI-TaqI.
