南方红豆杉茎段组织培养

Tissue Culture of Stem Segments of Taxus Chinensis Var. Mairei

摘要: 采用带芽嫩枝茎段为外植体直接诱导植株,研究南方红豆杉组织培养条件。结果表明: DCR+6-BA 0.5mg/L+NAA 0.8mg/L适合诱导南方红豆杉的芽; 1/2MS+NAA 0.1mg/L可以进行南方红豆杉根的诱导。将诱导出高约2cm健壮小苗转入DCR+NAA 0.1mg/L培养基诱导生根,45d左右有根长出,待根长约0.5 cm时,移栽至泥炭细沙混合培养基质中,在温度25℃、65%以上湿度下栽培30d左右,逐渐加大光照强度,植株成活率可达57.3%。

Abstract: Conditions of tissue culture of Taxus chinensis var.Softwood was studied by inducing stem segment with a axillary buds bud to plants directly. The results showed that the medium DCR +6- BA 0.5mg / L + NAA 0.8mg / L was most optimal bud-induced and the medium 1/2MS + N AA 0.1mg / L was root.-induced for Taxus chinensis var.Softwood. Robust seedlings at height about 2cm were moved into the medium DCR + NAA 0.1mg / L to root. Roots grew out about 45 days. when the root length of about 0.5 cm, the seedlings were transplanted into the peat and sand matrix to culture under temperatures 25 ℃, humidity above 70% in 30 days, gradually increasing light intensity, the plant survival rate reached to 57.3%.
