燕麦D基因组特异标记开发

Development of D Ggenome-specific Marker in Oat

摘要: 为了提高燕麦分子育种的效率,旨在开发燕麦D基因组特异分子标记。先利用300 条10 碱基随机引物对9 种燕麦种材料进行随机多态性扩增(random amplified polymorphic DNA,RAPD),扩增出燕麦D基因组特异性条带,命名为OPAB011287,经Blast 比对后发现该条带为一反转座子重复序列,根据此条带序列设计特异性引物,建立更简便稳定的SCAR标记。结果发现该SCAR标记可以准确特异的检测到9 种试验材料中含有D基因组的燕麦品种,结论表明该标记可用于燕麦分子标记辅助育种,有效服务于燕麦优良品种培育。

Abstract: The research aims to improve the efficiency of molecular breeding and develop D- Genome-specific markers in oat. Firstly, random amplified polymorphic DNA (RAPD) analysis was performed on 9 oat varieties with 300 random 10-based primers. A repetitive sequence of 1287 bp, named OPAB011287 was identified in the D genome of oat. BLAST search revealed that the sequence of OPAB011287 was highly homologous to a part of a retrotransposon. Based on the sequence of OPAB011287, a pair of sequence- characterized amplified region primers (SCAR) was designed which was more convenient and stable. And the results showed that SCAR marker was specific to target oat with D genome, which provided a powerful tool for oat marker assisted breeding and the better service for the breeding of oat.
