不同品种甘薯茎尖脱毒快繁技术优化研究

Research on virus-free and rapid propagation technology of sweet potato meristem culture

摘要: 为了建立更为有效的甘薯茎尖离体脱毒培养及快速繁殖技术体系,选用不同品种的甘薯对外源激素浓度、初次继代转培时间、自来水替代蒸馏水、白糖替代蔗糖等方面进行了试验研究。研究结果表明,以添加外源激素6-BA 0.5~1.0 mg/L和NAA 0.05~0.2 mg/L的MS培养基为最佳诱导分化培养基;茎尖接种后13~20天进行初次继代转培为最佳时间,成苗率最高达74.3%;继代快繁过程中用白糖替代蔗糖、自来水替代蒸馏水,成苗率可达到90%;脱毒苗在生产试验中优势明显,增产幅度为5.2%~68.6%。

Abstract: Different varieties of sweet potato were chosen to establish the virus-free and rapid propagation of meristem culture technology system more effectively, the experiments included hormone concentration research, the first time of transgenic subculture, distilled water was replaced by running water, sucrose was replaced by white granulated sugar and so on. The results showed that the MS medium contains 0.5-1.0 mg/L 6-BA, 0.05-0.2 mg/L NAA was the optimal medium for differentiation; 13-20 days after transplantation was the optimal time for transgenic subculture, the survival rate was as high as 74.3%; there was a high seedling rate (90%) when replaced sucrose and distilled water by white granulated sugar and running water; virus-free seed had obvious advantages in production, had a yield increase ranging from 5.2%~68.6%.
